利用HPLC-ICP-MS方法测定中华绒螯蟹不同形态砷的残留量 总被引:1,自引:1,他引:0
采用高效液相色谱串联电感耦合等离子体质谱仪(HPLC-ICP-MS)测定中华绒螯蟹可食部位6种砷形态的残留量。经过前处理、提取和脱脂后,通过色谱柱、流动相、pH和流速等参数调节,筛选得到最优化的分离和检测条件。结果表明,6种砷形态的混合标准溶液在质量浓度0~50 μg/L范围时,均呈现良好的线性关系,其R2达到0.999,检测限为0.0060~0.0912 μg/kg,定量限为0.020~0.304 μg/kg。在样品中添加0.5、5、50 μg/L浓度水平,回收率良好,范围是70%~120%,相对标准偏差(RSD)均<5%,方法精密度与重复性均良好。本方法为中华绒螯蟹可食部位砷形态残留量建立了有效检测方法,为水产品砷残留快速检测提供技术支撑。 相似文献

This study was conducted to explore the multiplication pattern of the recombinant strain Re-7 of avian influenza virus subtype H5 in Madin Darby Canine Kidney(MDCK) cells and to determine the optimal multiplicity of infection(MOI) and the optimal time for virus harvest. The recombinant strain Re-7 was inoculated at different MOIs into MDCK cells grown in serum-free medium in 100 L bioreactors for replication. Then,the hemagglutination(HA) titer, 50% tissue culture infectious dose(TCID50) and 50% embryo infectious dose(EID50) of culture medium were measured once every 12 h from 24 h after virus inoculation to determine the optimal MOI. After that, virus was inoculated at the optimal MOI deter-mined above into MDCK cells for large-scale virus replication to determine the optimal time for virus harvest. The results showed that the optimal MOI was 10~(-2), and the optimal time for virus harvest was 60 h after inoculation. Under these conditions, the HA titer, TCID_(50) per 1 m L and EID50 per 0.1 m L were increased to 1∶102 4, 10~(7.33) and 10~(6.83), respectively. This study aimed to determine the optimal protein to energy ratio (P/E ratio) and evaluate the effect of dietary protein and lipid levels on growth performance, body composition and digestive enzymes activities in Chinese mitten‐handed crab, Eriocheir sinensis. Nine practical diets containing three levels both for protein (DP 30%, 35% and 40%) and lipid (DL 2%, 7% and 12%) with P/E ratios ranging from 13.69 to 19.79 mg KJ?1 were fed to four replicates of crabs (3.39 ± 0.10 g) for 10 weeks. Weight gain increased significantly with the increase in DP level at each DL level. Moreover, weight gain increased in crabs fed with diets containing DL level from 2% to 12% and DP level from 30% to 35%. However, the diet containing 40% DP and 12% DL levels significantly decreased the growth performance and protein efficiency of the crabs. The whole crab and hepatopancreas lipid contents also increased as dietary lipid increased, but not dietary protein. The total protease activity increased significantly with the increase in dietary protein at each lipid level. The lipase activity was statistically comparable among different DL levels at each DP level. Taken together, the crab fed the diet containing 35% protein and 12% lipid levels with P/E 15.77 mg KJ?1 revealed optimal growth, feed utilization efficiency and digestive enzymes activities. Moreover, our study indicated that the higher dietary lipid level at a relatively lower dietary protein level could provide protein sparing effect in Eriocheir sinensis. 相似文献

White spot syndrome virus (WSSV), an enveloped double‐stranded DNA virus, is the causative agent of a disease that has led to severe mortalities of cultured shrimps in Taiwan and many other countries. In the previous study, Penaeus monodon chitin‐binding protein (CBP) and glucose transporter 1 (Glut1), two cell membrane proteins, were found to at least interact with other 10 WSSV envelope proteins including VP51B. These envelope proteins might form a protein complex. According to the known information, VP51B was used to identify its role in the protein complex. Western blotting of the intact viral particles and fractionation of the viral components confirmed that VP51B is one of WSSV envelope proteins. In this study, the protein–protein interaction between VP51B and other WSSV envelope proteins was identified by far‐western blot experiment and VP51B was found to interact with VP24, VP31, VP32, VP39B and VP41A. Furthermore, the in vivo neutralization experiment using recombinant VP51B plus with VP39B showed the best inhibition. These data indicate that VP51B participates in the WSSV protein complex and plays an important role in WSSV infection. 相似文献

Indoor and outdoor experiments were carried out simultaneously to evaluate the effects of dietary phosphorus levels on growth performance and body composition of crucian carp. Monocalcium was used to formulate extruded feed containing available phosphorus (AP) of 1.5 (D1), 4.7 (D2), 6.3 (D3) and 9.0 (D4) g kg?1. Twelve 500‐L recirculating tanks were assigned to four triplicate groups with each tank stocked with 15 juveniles (39.88 ± 0.14 g) for indoor experiment. Twelve polyethylene enclosures were assigned to four groups with each one stocked with 120 juveniles (44.08 ± 1.36 g). After 8 weeks of feeding, survival ranged from 96% to 100%. Weight gain, special growth rate, protein efficiency ratio and feed conversion rate did not show significant differences among dietary treatments in both experiments. Fish cultured in the enclosures grew better, although D1 and D2 treatments did not reach significant level. Crude protein and phosphorus content of the fish were not significantly affected by dietary phosphorus level. Moisture, crude lipid, crude ash and vertebrae phosphorus were significantly affected by different phosphorus levels. Based on vertebrae phosphorus, crucian carp cultured in the tanks and in the enclosures showed phosphorus deficiency under 4.7 g kg?1 and 6.3 g kg?1 AP, respectively. 相似文献

Acid soluble collagen was extracted from the scales of lizardfish and characterized with sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and amino acid analysis. After 8 h of collagen hydrolysis, hydrolysates had an estimated degree of hydrolysis (DH) from 4 ± 0.05 to 25 ± 0.63% (p < 0.05). All hydrolysates had angiotensin converting enzyme inhibitory and antioxidant activity. These activity levels showed little change after treatment with gastrointestinal proteases. Results indicated that the lizardfish by-products may be improved by enzymatic treatment with acid-soluble collagen from lizardfish scales. 相似文献

A feeding trial was conducted to evaluate the effects of replacing fish meal with rubber seed meal (RSM) on growth, nutrient utilization, and cholesterol metabolism of tilapia (Oreochromis niloticus × Oreochromis aureus). Five experimental diets were formulated with 0, 150, 300, 450, and 600 g kg?1 RSM replacing graded levels of fish meal, respectively. Each diet was randomly assigned to triplicate groups of 25 fish (initial average weight 65.3 g) per aquarium in a rearing system maintained at 29 ± 1 °C for 8 weeks. Dietary 150 g kg?1 RSM inclusion did not affect the weight gain and daily growth coefficient, whereas these were depressed by a further inclusion. Additionally, feed efficiency ratio and protein efficiency ratio were not affected by dietary RSM inclusion regardless of inclusion level. However, the inclusion of 450 and 600 g kg?1 RSM decreased the mid-intestinal trypsin, lipase, and amylase activities; the hepatic acyl-CoA/cholesterol acyl transferase; low-density lipoprotein receptor; and 3-hydroxy-3-methyl-glutaryl-CoA reductase activities. Similarly, dietary 600 g kg?1 RSM inclusion inhibited the plasma catalase and hepatic glutathione peroxidase activities. These results indicated that 150 g kg?1 RSM can be included in tilapia diets, whereas higher inclusion of RSM inhibited the growth rate, digestive enzyme activity, antioxidant capacity, and cholesterol metabolism. 相似文献
